The effect of hydrogen-rich water on letrozole-induced polycystic ovary syndrome in rats.

Abstract

This animal study examined how hydrogen-rich water affects a rat model of polycystic ovary syndrome (PCOS) induced by letrozole administration. Female rats were allocated to four groups: control (carboxymethyl cellulose solution), H2 alone (hydrogen-rich water plus CMC), PCOS (letrozole in CMC), and PCOS plus H2 (letrozole plus hydrogen-rich water). Biochemical and histopathological analyses were performed on collected blood and ovarian tissue. Ovarian cyst counts were significantly lower in the PCOS plus H2 group than in the PCOS group (P < 0.0001). Markers of oxidative stress and hormonal imbalance — malondialdehyde, cortisol, and testosterone — were all significantly reduced in the hydrogen-treated PCOS animals (P = 0.0458, 0.0003, and 0.0041, respectively), while glutathione levels rose significantly (P = 0.0012). These findings indicate that hydrogen-rich water can attenuate both cyst formation and oxidative damage in letrozole-induced PCOS.

Mechanism

Hydrogen-rich water is proposed to scavenge reactive oxygen species, thereby reducing lipid peroxidation (malondialdehyde) and restoring glutathione levels, while also suppressing excess cortisol and testosterone secretion, collectively limiting ovarian cyst development in PCOS.

Hydrogen-rich water is a low-risk delivery route, but the achievable systemic hydrogen dose is bounded. For clinical applications, inhalation is the most efficient route; inhalation, however, carries explosion risk, and concentration matters (empirical LFL of 10% applies to inhalation environments; high-concentration devices are documented in the Consumer Affairs Agency accident database and are not recommended).
